Synthesis of New Collision-Induced Cleavable Photoactivatable Protein Cross-Linkers

Abstract
The following document describes the completed synthetic route to heterobifunctional lysine-photoactivatable linker succinimide benzophenone sulfoxide (SBPSO) and a partially completed route to a trifluoromethylphenyldiazirine (TPD) photoactivatable cross-linker. In order to expand on photoreactive cross-linking chemistry, a new class of aryl azide heterobifunctional lysine-photoactivatable cross-linkers was also developed. Four aryl azides: succinimide para-azido sulfoxide (p-SAzSO), succinimide tolyl-azido sulfoxide (STAzSO), succinimide para-azido meta-methyl sulfoxide(p-SAzMSO), and succinimide meta-azido sulfoxide (m-SAzSO) were synthesized. All synthesized linkers will be used for in vitro and in vivo protein interaction elucidation studies as well as protein structural dynamic probing studies analyzed through liquid chromatography tandem mass spectrometry.
